Abstract:
Objective:To explore the role and regulatory mechanism of taurine upregulated gene 1 (TUG1) in the invasion and metastasis of gastric cancer cells.Methods:TCGA database analysed TUG1 expression in gastric cancer tissue and its relationship with patient prognosis.TUG1 expression in gastric cancer tissues and cells was detected by qRT-PCR.TUG1 overexpressed knockdown cell line was constructed by cell transfection.Transwell experiment observed the effects of TUG1 overexpression and knockdown on the migration and invasion ability of gastric cancer SGC-7901 cells.Western Blot detected the effects of TUG1 overexpression and knockdown on the activity of the EZH2/STAT3 signaling pathway in gastric cancer SGC-7901 cells.Results:qRT-PCR and TCGA database showed that TUG1 expression was up-regulated in gastric cancer tissues and cells (P<0.01),and was negatively correlated with prognosis of gastric cancer patients (P<0.01),but positively correlated with pathological stages and lymphatic metastasis of gastric cancer patients (P<0.05).Transwell assay showed that upregulation of TUG1 promoted the migration and invasion of gastric cancer cells (P<0.01).Western Blot result showed that upregulation of TUG1 promoted EZH2 expression and activated STAT3 signaling pathway (P<0.01).Conclusion:TUG1 promotes the invasion and metastasis of gastric cancer cells by activating EZH2/STAT3 signaling pathway.
