Abstract:
Objective:To investigate the role of silybin in inhibiting proliferation and promoting apoptosis of breast cancer MCF7 cells.Methods:The cells were divided into control group and SIL groups (10,20,40mg/L),each group was treated for 24,48 and 72h.MTT method was used to detect the survival rate of cells.TUNEL was used to detect the apoptotic rate of cells.Special kits were used to detect the activity of caspase3/7.Western bolt method was used to detect the expression of PUMA.Results:The MTT results show that SIL could inhibit the proliferation of MCF7 cells on dose and time-depend manner,the effect of 40mg/L SIL (72h) was the most obvious.After being treated with SIL (48h),the apoptotic rate of cells and the activity of caspase3/7 increased significantly.In addition,PUMA protein increased significantly in SIL treated group.Conclusion:Silybin can inhibit proliferation and promote apoptosis of breast cancer MCF7 cells.PUMA may play a role in the apoptosis of breast cancer MCF7 cells induced by silybin.
