Abstract:
Objective:To investigate the effect of quercetin on the proliferation of oral squamous cell carcinoma cells by regulating the stability of p21/cip1 and p27/kip1 proteins.Methods:CCK-8 kit was used to detect the proliferation of oral squamous cell carcinoma cells (SCC-15).Flow cytometry was used to detect the cell cycle distribution of SCC-15.Western blot was used to detect CyclinD1/CDK complex,p21/cip1,p27/kip1,p-GSK3β,GSK3β,p53 and c-Myc protein expression.qRT-PCR was used to detect the mRNA expression of p21/cip1 and p27/kip1 in SCC-15 cells.Laser confocal microscope was used to detect the subcellular localization of p21/cip1 and p27/kip1 protein in SCC-15 cells.Cycloheximide chase assay was used to detect p21/cip1 and p27/kip1 protein half-life in SCC-15 cells.Results:Compared with the control group,quercetin inhibited the proliferation of SCC-15 cells in a dose-dependent manner (6.25 μmol/L,12.5 μmol/L,25 μmol/L,50 μmol/L,100 μmol/L,and 200 μmol/L)(P<0.05).IC50 of quercetin on SCC-15 cells at 48 h was 80.07 μmol/L.Compared with the control group,quercetin (40 μmol/L and 80 μmol/L) can significantly increase the proportion of G1/G0-phase cells in SCC-15 cells,significantly reduce the proportion of S-phase and G2/M-phase cells (P<0.05) in a dose-dependent manner,inhibit the expression of CyclinD1/CDK complex,p-GSK3β and c-Myc protein (P<0.05),promote the expression of mRNA and protein of p21/cip1 and p27/kip1,and promote the expression of GSK3β and p53 protein (P<0.05).Compared with the control group,quercetin (80 μmol/L) promoted the transfer of p21/cip1 protein from cytoplasm to nucleus in SCC-15 cells (P<0.05),and significantly increased the biological half-life of p21/cip1 and p27/kip1 proteins (P<0.05),no significant effect on the subcellular localization of p27/kip1 protein (P>0.05).Conclusion:Quercetin inhibits the proliferation of oral squamous cell carcinoma by regulating the stability of p21/cip1 and p27/kip1 proteins.
