Abstract:
Objective:To study the expressions of microRNA-145(miR-145) and KLF5 in bone marrow fluid of multiple myeloma and their clinical significances.Methods:From January 2015 to March 2016,65 patients with multiple myeloma(MM) and 36 healthy subjects(control group) were selected as subjects.Real-time fluorescence quantitative polymerase chain reaction(qRT-PCR) was used to detect the expression levels of miR-145 and KLF5 mRNA.Western Blot was used to detect the expression of KLF5 protein.The correlation between the expressions of miR-145 and KLF5 in multiple myeloma bone marrow fluid was analysed.Results:The expression of miR-145 in MM bone marrow fluid was significantly lower than that in normal control group(P<0.05).The expressions of KLF5 mRNA and protein were significantly higher than those in the control group(P<0.05).Compared with phase I and phase Ⅱ,the expression of miR-145 in bone marrow fluid of phase Ⅲ was significantly decreased(P<0.05),and the expression of KLF5 mRNA was significantly increased(P<0.05).Compared with IgG and IgA,the relative expression level of miR-145 in bone marrow fluid of light chain and non-secretory patients was significantly lower(P<0.05).There was no significant difference in the relative expression level of KLF5 in different immunophenotypes(P>0.05).The expression of miR-145 was negatively correlated with that of KLF5 mRNA(r=-0.452,P<0.05).The expression of miR-145 in bone marrow fluid of dead MM patients was significantly lower than that of survivors(P<0.05),and the expression of KLF5 was significantly higher than that of survival patients(P<0.05).Conclusion:The expression of miR-145 in bone marrow fluid of patients with MM is low,KLF5 is highly expressed,which may be closely related to the occurrence and development of multiple myeloma.
