Abstract:
Objective:To investigate the relationship between lipid-metabolism related gene SCARB1 and RCC and its potential biological effect.Methods:The expression of SCARB1 mRNA in normal kidney and RCC tissues in TCGA database was analyzed,and immunohistochemistry was used to detect the level of SCARB1 protein in RCC TMA set.Furthermore,SCARB1 DNA copy number alteration and methylation were analyzed from TCGA database.Kaplan-Meier analysis was performed to detect the relationship between SCARB1 level and overall survival of RCC patients.Moreover,Oil Red O staining was used to detect the lipid level in SCARB1 low and high tissues,and the association between SCARB1 and a lipid-regulation gene SCD was analyzed by Pearson's correlation analysis.Results:The expression of SCARB1 mRNA and protein were significantly increased in RCC tissues compared to normal kidney.A gain of SCARB1 DNA copy number was observed in RCC tissues compared to normal kidney,and a negative association was observed between SCARB1 mRNA expression and methylation.Furthermore,SCARB1 highly expressed RCC patients showed poor overall survival,and there was a positive correlation between SCARB1 and SCD levels.Conclusion:SCARB1 is increased in RCC and indicates a poor prognosis of patients.Furthermore,SCARB1 may play critical role in the lipid synthesis of RCC cells.
