Abstract:
Objective:To evaluate the expression of lncRNA XIST in esophagus cancer tissues and investigate the effect of lncRNA XIST on the proliferation of esophagus cancer cells.Methods:The expression of lncRNA XIST in 23 pairs of esophagus cancer and adjacent tissue were determined by real-time PCR.The expression of miR-486-5p in three esophagus cancer cell lines was also detected.The two lncRNA XIST-high-expressed cells were transfected with lncRNA XIST siRNA or NC siRNA and the interfere efficient was determined by qRT-PCR.Cell proliferation activities were examined by CCK-8 assay.Dual-Luciferase reporter assay was also used to exam whether miR-486-5p could bind to lncRNA XIST.Moreover,the expression of miR-486-5p was also detected by qRT-PCR.Results:The expression of lncRNA XIST was significantly increased in esophagus cancer tissues compared with cancer-adjacent tissue (P<0.01).Eca-109 and TE-1 cells showed the highest level of lncRNA XIST in the three esophagus cancer cell lines.Transfect with lncRNA XIST siRNA in Eca-109 and TE-1 cells reduced the level of lncRNA XIST to (75.50±0.89)% and (64.74±13.42)% respectively.Cell proliferation activity of Eca-109 and TE-1 cells was also significantly reduced by lncRNA XIST siRNA.Dual-Luciferase reporter assay showed that the relative luciferase activity was significantly increased in wt-XIST +miR-486-5p mimic group.Furthermore,the expression of miR-486-5p was also up-regulated by lncRNA XIST siRNA.Conclusion:lncRNA XIST is closely related to the occurrence and development of esophagus cancer,miR-486-5p may be a novel target in the prevention and treatment of esophagus cancer.
