Abstract:
Objective:To study the effects of silencing variant plasmacytoma heterotopic 1 (PVT1) gene on proliferation,migration,signal transduction and activation factor (STATs) 3 pathway of ovarian cancer SKOV3 cells.Methods:SKOV3 cell line of ovarian cancer was cultured in vitro.The sequence of PVT1 siRNA cell was designed,cells were transfected.The study in cluded siRNA group,negative control group (NC) and blank control group (BC).The relative expression of PVT1 was detected by qRT-PCR.MTT assay was used to detect cell proliferation.Scratch test was used to detect cell migration ability.Transwell assay was used to detect cell invasiveness.The expression of STAT3 pathway-related protein was detected by WB method.Results:After transfection,the expression of PVT1 in cells in siRNA3 group was significantly lower than that in BC and NC groups (P<0.05).After transfection,compared with BC group and NC group,the cell proliferation rate of siRNA3 group was significantly lower than that of BC group and NC group (P<0.05),the numbers of migrating and invading cells decreased significantly (P<0.05),the expression levels of p-STAT3 protein,MMP2,MMP9,CD44 and PCNA were significantly decreased (P<0.05).Conclusion:Silencing PVT1 may inhibit the proliferation,migration and invasion of ovarian cancer SKOV3 cells,and its specific mechanism may be related to STAT3 signaling pathway.
