Abstract:
Objective:To investigate the molecular mechanism of Ghrelin induces breast cancer cell MDA-MB-231 proliferation.Methods:Human breast cancer cell MDA-MB-231 were treated with Ghrelin,[D-Lys3]-GHRP-6 and Rapamycin.The proliferation of MDA-MB-231 was determined by MTT assay and BrdU assay.The expression of GHSR and phosphorylation of mTOR,p70S6K,S6 was detected by Western blot.Results:The proliferation of MDA-MB-231 cells was increased by Ghrelin.Western blot showed that GHSR was expressed on MDA-MB-231 cells and Ghrelin induced rapid phosphorylation of mTOR,p70S6K and S6,which was lower in D-L-GHRP group compared with the Ghrelin group.Conclusion:The results demonstrate that Ghrelin induces MDA-MB-231 cells proliferation through binding to GHSR and activating mTOR/p70S6K/S6 signaling pathway.
