Abstract:
Objective:To investigate the effects of miR-301b and miR-769-5p on cell growth in non-small cell lung cancer A549 cell line.Methods:A549 cells were transfected by lentiviral expression vector of miR-301b and miR-769-5p gene.Cell cycle and apoptosis were detected by flow cytometry and proliferation were detected by MTT assay after transfection.Results:The result of qRT-PCR showed that miR-301b and miR-769-5p expression in A549 cell infected by miR-301b and miR-769-5p were higher than others(P<0.01).G0/G1 phase cycle was increased,S and G2/M phase cycle were arrested after up-regulation of miR-301b,S phase was arrested,G0/G1 and G2/M phase cycle were increased after up-regulation of miR-769-5p in A549 cell.Conclusion:miR-301b was validated to regulate FOXF2,and miR-769-5p modulated ARID1A.Additionally,overexpression of miR-301b and miR-769-5p affected cell cycle of A549 cells,but not cell proliferation and apoptosis.It may be helpful for further investigation of the regulation of the target effect for the molecular target therapy of lung cancer.
