Abstract:
Objective:To investigate the expression level of E2F1 and its physiological significance in vulvar squamous cell carcinoma(VSCC).Methods:Semi-quantitative RT-PCR,Western blot and immunohistochemistry were used to analyze E2F1 mRNA and protein expressions in 30 cases of VSCC and adjacent normal tissues.Clinical records and the pathology of the frozen specimens were collected to analyze the relationship between E2F1 and clinical pathology.Results:Semi-quantitative RT-PCR showed the average E2F1 mRNA expression levels were (1.27±0.04) and (0.84±0.06) in VSCC tissues and adjacent normal tissues(P<0.05).Western blot showed average E2F1 protein expression levels were (1.16±0.04) and (0.93±0.03) in VSCC tissues and adjacent normal tissues(P<0.05).Immunohistochemistry showed the protein positive expression rate of E2F1 in VSCC tissues (56.67%,17/30) was significantly higher in VSCC than that in normal tissues (33.33%,10/30) (P<0.05).The mRNA positive expression rate of E2F1 was positively correlated with FIGO stage.Conclusion:The up-regulation of E2F1 in VSCC was found to be associated with FIGO stage,E2F1 might be associated with the tumourigenesis and development of VSCC.
