Abstract:
Objective:To investigate the effects of miR-300 on the proliferation,apoptosis,and cell cycle of humanovarian cancer cells.Methods:The expression of miR-300 in the normal ovarian epithelial cell and ovarian cancer cells,and the silencing efficiency of miR-300 siRNA was detected by quantitative Real time PCR(qRT-PCR).After siRNA and NC transfected into ovarian cancer cells for 48 hours,the proliferation of ovarian cancer cells were analyzed by CCK-8,the apoptosis and cell cycle of ovarian cancer cells was analyzed by flow cytometry.Results:The expression of miR-300 was significantly upregulated in ovarian cancer cells comparing with normal ovarian epithelial cells and could be effectively silenced by miR-300 siRNA.Proliferation of ovarian cancer cells were inhibited after transfected miR-300 siRNA,but cells apoptosis were promoted.Cell cycle of ovarian cancer cells were arrested at stage of G1 after transfected miR-300 siRNA.Conclusion:miR-300 play an important role on the proliferation,apoptosis and cell cycle of ovarian cancer cells.
