Abstract:
Objective:To investigate the effect and mechannisms of TSPAN8 gene on cell proliferation of A549.Methods:Human lung cancer cells A549 was transfected with pCMV-TSPAN8 plasmid.The proliferation of A549 cells was determined by MTT assay,flow cytometry was applied to detect the impact of TSPAN8 over-expression on cell cycle.The effects of TSPAN8 gene on expression on Rb1,E2F1,C-myc,CDK4 and Cyclin D1,and the phosphorylation of ERK in the A549 cells were determined using Real-time PCR and Western blot assay.Results:TSPAN8 gene could promote cell proliferation of human lung cancer cells in vitro.Western blot and Real-time PCR showed that protein and mRNA expression of E2F1,C-myc,CDK4 and Cyclin D1 were up-regulated after transfected with TSPAN8 gene in the A549 cells,but expression of Rb1 was down-regulated.At the same time,the phosphorylation of ERK was also activated as well.Conclusion:TSPAN8 gene could promote cell proliferation and metastasis of human lung cancer cells,and its effect might through facilitation of expression of Rb1,E2F1,C-myc,CDK4 and Cyclin D1,and activation of the ERK1/2 signal pathway.
