Abstract:
Objective:To study the function of miRNA-9 on osteosarcoma MG-63 cells proliferation and apoptosis and correlated mechanisms by improving the expression of miRNA-9.Methods:Osteosarcoma MG-63 cells were randomly divided into three groups:Experimental group(transfected with miRNA-9 Oligo),control group(transfected with miRNA negative contrat)and blank group(transfected nothing).Detected the expression of miRNA-9,CXCR4 by real time PCR.Detected the CCK value(OD)at 24h,48h,72h and 96h,then observed the cell growth curve.The apoptosis was measured by flow cytometry.Results:Compared with negative control group and blank group,the miRNA transfection group was higher expressed on miRNA-9,osteosarcoma cells showed lower cell proliferation efficiency and higher cell apoptosis rotio on miRNA-9 higher expression groups.Conclusion:CXCR4 gene involved in the miRNA-9 inhition of osteosarcoma MG-63 cell proliferation,and promote cell apoptosis.
