Abstract:
Objective:To explore the high-intensity focused ultrasound(HIFU)in treatment of in vitro PANC-1 pancreatic cancer cell lines,then observe still alive cells of STAT3/pSTAT3 protein expression and the change of cell apoptosis.Methods:To establish in vitro agarose gel model embedded human pancreatic tissue,then were randomly divided into control and experimental group and tagged.Experimental group was treated by type JC200 HIFU therapeutic equipment,control group without any treatment.The specimen pathological changes were observed by HE dyeing and the STAT3/pSTAT3 protein expression changes were observed by immunohistochemistry.In addition,in vitro experimental model of pancreatic cancer cell lines were respectively irradiated with different doses of HIFU (0J,100J,200J,400J,800J).After that,observing pancreatic cancer cell proliferation inhibition by MTT,and detecting STAT3/pSTAT3 expression by Western blot.Results:After HIFU treatment,in vitro of pancreatic cancer target tissues histopathological obviously observed solidified necrosis,and cell structure disappeared.Immunohistochemical staining observed target tissues brown granules decreased significantly,which demonstrated STAT3/pSTAT3 protein expression decreased after treatment.MTT cell proliferation rate demonstrated control group (0J) compared to low-power groups (100J,200J,400J) had no significant differences in cell proliferation,and high-power group (800J) significantly reduced the rate of cell proliferation,no significant difference between groups(P>0.05).Western blot tested expression of STAT3/pSTAT3 suggested low-power groups (100J,200J,400J) had no significant differences with the control group.Conclusion:HIFU can effectively treat pancreatic cancer,which may be related to destroy tumor cells themselves structure and STAT3/pSTAT3 protein expression.
