Abstract:
Objective:To detect the expression of GRK6 in bile duct carcinoma tissues and adjacent normal bile duct epithelial tissues,and to explore the effects of inhibiting GRK6 expression on the proliferation and invasion of bile duct carcinoma cells.Methods:The expression of GRK6 in cholangiocarcinoma and adjacent normal tissues was detected by qRT-PCR,immunohistochemical staining,and the relationship between GRK6 expression and different clinicopathological factors was analyzed.GRK6 expression in QBC939 cells was inhibited by RNA interference technique,and the inhibitory effect of siRNA on GRK6 in QBC939 cells was detected.The effects of GRK6 inhibition on the proliferation of bile duct cancer cells were detected by MTT and plate clone formation assay,and the effects of GRK6 inhibition on cell invasion were detected by Transwell assay.Results:Both qRT-PCR and immunohistochemical results indicated that GRK6 was highly expressed in cholangiocarcinoma tissues.The high expression of GRK6 was closely related to tumor size and TNM stage.si-GRK6 effectively silenced the expression of GRK6 in QBC939 cells.The results of MTT assay and plate clonogenesis assay showed that inhibition of GRK6 expression could inhibit the proliferation and clonogenesis of bile duct carcinoma cells QBC939.Transwell assay results showed that inhibition of GRK6 expression significantly reduced the invasion ability of bile duct cancer cells.Conclusion:GRK6 is highly expressed in bile duct cancer cells.Inhibition of GRK6 expression can inhibit the proliferation,invasion of bile duct cancer cells.
