Abstract:
Objective:To investigate the role of hsa-miR-196a-5p in the occurrence and development of human esophageal squamous cell carcinoma,and to predict its biological characteristics and functions,and to provide useful clues for the final study of the function of miR-196a-5p.Methods:The relative expression levels of miR-196a-5p in paraffin tissue and adjacent tissues of patients with esophageal squamous cell carcinoma were detected by Quantitative Real-time PCR.MIRDB,miRanda,Target Scan and Tarbase DIANA TOOLS 7.0 were used to predict the targeted regulatory genes of miR-196a-5p,and the online software VENNY2.1 was used to obtain the intersection to finally determine the target gene collection range of miR-196a-5p.The online database string query is used to query the correlation between the predicted target gene encoded proteins.Finally,we used the online database metascape to perform functional annotation classification and pathway enrichment analysis based on GO and KEGG databases for the target gene set predicted by hsa-miR-196a-5p.Results:miR-196a-5p was located in human 12q13.13.Checking the mirBase database,miR-196a-5p and miR-196a-5p of zebrafish,lamprey,platypus,brown house mouse,rabbit,wild boar and other species were highly conserved in the UAGGUAGUUUCAUGUUGUUGGG region.The expression of miR-196a-5p in 32 pairs of esophageal squamous cell carcinoma tissues was significantly higher than that in the corresponding adjacent tissues,which was consistent with the results of the previous Agilent Human miRNA gene chip.The final target genes were 48,and the relationship networks between the 48 target genes encode proteins concentrated in the HOX family with HOXA5,HOXA7 and HOXA9 as the core and two major groups with EXOC8 as the core.The functions of miR-196a-5p target genes were concentrated in embryo and organ morphogenesis,development of skeletal system,transcriptional regulation of RNA polymerase Ⅱ promoter,positive regulation of gene expression/transcription,and participation in regulation of transcription factor activity.The main signal pathways involved were Ras,FoxO signal pathway,AGE-RAGE signal pathway,insulin signal pathway and tumor transcription disorder pathway.miR-196a-5p was highly expressed in ESCC tumor tissue,while TGFBR3 gene had the opposite expression pattern with miR-196a-5p.We speculated that TGFBR3 may be the target of miR-196a-5p in ESCC.Conclusion:miR-196a-5p is highly expressed in esophageal squamous cell carcinoma,suggesting that miR-196a-5p can be involved in the development of esophageal cancer as a cancer promoter.
