Abstract:
Objective:To investigate the effect of nano-hydroxyapatite (Nano-HAP) on proliferation and apoptosis of human osteosarcoma U2OS cells.Methods:Human osteosarcoma cells U2OS were treated with Nano-HAP at different concentrations.The effect of nano-HAP on cell adhesion was detected by FDA staining.And the effect of Nano-HAP on cell proliferation was detected by CCK8 assay and plate clone formation.At last,the AnnexinⅤ-FITCapoptosis detection kit was used to detect the effect of Nano-HAP on cell apoptosis.Results:Different concentrations of Nano-HAP had no significant effect on the adhesion of U2OS,but had a significant inhibitory effect on the cloning formation,meanwhile,the proliferation of U2OS cells could be significantly inhibited at 50 μg/ml and 800 μg/ml.Conclusion:Nano-HAP could significantly inhibit the proliferation of osteosarcoma cell U2OS and promote cell apoptosis,however,it has no significant effect on the initial cell adhesion.
