IL-17A promotes the proliferation,migration and invasion of lung cancer A549 cells through PI3K/Akt pathway

Objective:To study the effect and molecular mechanism of IL-17A on proliferation,migration and invasion of lung cancer A549 cells.Methods:A549 cells were treated with different concentrations of IL-17A(0,1,10,100 ng/mL) respectively.CCK-8 was used to detect the effect of IL-17A on the proliferation of A549 cells.Wound healing assay and Transwell migration assay were used to detect the effect of IL-17A on the migration and invasion of A549 cells.Western blot was used to detect the effect of IL-17A on the protein expression of PI3K/Akt signaling pathway and apoptosis pathway.Flow cytometry was used to detect the effect of IL-17A on A549 cell apoptosis.A549 cells were treated with IL-17A and PI3K inhibitor LY294002 together,wound healing assay was used to observe the effect on the migration of A549 cells.Results:IL-17A promoted the proliferation of A549 cells,the promoting effect increased with the increase of IL-17A concentration.IL-17A promoted the expression of PI3K and Akt phosphorylated proteins and inhibited the protein expression of caspase3,and inhibit the apoptosis of A549 cells.LY294002 could significantly reduce the effect of IL-17A on the migration of A549 cells.Conclusion:IL-17A inhibits the apoptosis of A549 cells through PI3K/Akt pathway,thereby promoting the proliferation,migration and invasion of A549 cells.
