Abstract:
Objective:To construct pancreatic cancer cell lines with lnc-TNFAIP3 gene overexpression by lentivirus transfection and observe the changes of cell behavior.Methods:The overexpressed plasmid and lentivirus were prepared and transfected into human pancreatic cancer cell line PANC-1.The cell proliferation was detected by CCK-8 and cloning assay,the migration and invasion were detected by scratch and Transwell,the protein expression was detected by Western blot,and the cell cycle and apoptosis were detected by flow cytometry.Results:PANC-1 were successfully transfected with lentivirus overexpressing lnc-TNFAIP3 gene,and the proliferation,migration and invasion of PANC-1 were decreased.There was no significant difference in cell cycle and apoptosis,and no significant difference in the expression of related proteins.Conclusion:Human pancreatic cancer cell line PANC-1 constructed by lentivirus transfection with lnc-TNFAIP3 gene overexpression has changed in cell behavior,the present results suggest that lnc-TNFAIP3 may be a promising therapeutic target for pancreatic cancer,which provide a theoretical and experimental data for the study of lnc-TNFAIP3 gene in pancreatic cancer.
