Abstract:
Objective:To explore the expression of urine exosomes in urothelial cell carcinoma of the bladder and its clinical relevance.Methods:Tumour-derived exosomes in urine samples of 30 urothelial cell carcinoma of the bladder patients and 15 healthy control peoples were separated using ultracentrifugation.Morphologic characteristics of exosomes were observed by the electron microscope and protein concentration was determined by the BCA protein assay kit.The expression of CD9 in exosomes was detected by Western blot.Results:The level of urine exosomes in healthy control people was significantly different from that in experimental group [(125.99±47.71)μg/μL vs (259.74±57.47)μg/μL,P<0.000 1].According to the degree of tumor invasion,there was a statistically significant difference in urine exosome level between patients with non-muscle invasive bladder cancer and patients with muscle-invasive bladder cancer.However,muscle-invasive bladder cancer patients were no significant differences between subgroups.Conclusion:Detection of urine exosome may offer a convenient tool for early diagnosis of urothelial cell carcinoma of the bladder.
