Abstract:
Objective:To investigate the clinical significance of miR-455-3p expression in colorectal cancer(CRC) tissues and its effect on cell proliferation.Methods:Based on the data from the GEO platform(GSE30454 and GSE108153),the limma package was used to analyze the significantly differentially expressed miRNAs in CRC tissues.The expression difference of miR-455-3p in CRC tissues was analyzed by UALCAN platform containing TCGAdatabase,and the effect of miR-455-3p on survival was preliminarily assessed by Kaplan-Meier Plotter website.Besides,in situ hybridization(ISH) was performed to detect miR-455-3p expression in CRC and matched adjacent tissues(n=185),and the dysregulation was further compared by Wilcoxon rank sum test.Furthermore,the correlation between miR-455-3p expression and clinicopathological parameters was revealed using Pearson χ2 test and unconditional Logistic regression.The potential of miR-455-3p expression in predicting the prognosis of patients with CRC was evaluated by Kaplan-Meier based on Log-rank test,multivariate COX regression,and multifactor dimensionality reduction(MDR) method.And finally,the expression of miR-455-3p in CRC cell lines and its effect on cell proliferation was detected by qPCR and MTT assays.Results:miR-455-3p,miR-1181,miR-1290,and miR-622 were the co-dysregulated miRNAs in the two GEO microarrays,among which miR-455-3p was dramatically down-regulated in CRC tissues and closely associated with poor prognosis of patients(P=0.007 9,HR=0.21) based on TCGA database.Then,the decreased miR-455-3p level in CRC was significantly correlated with tumor size and primary site(P<0.05).High miR-455-3p expression predicted prolonged survival time of CRC patients(P<0.001),and acted as an independent protective factor for prognosis(P<0.001,HR<1).More importantly,the best model composed of miR-455-3p-univariate showed the compelling potential to effectively evaluate disease-free survival(DFS) and overall survival(OS)(P<0.05,Cross-validation consistency=10/10).miR-455-3p was reduced in CRC cell lines(P<0.05),and its overexpression significantly inhibited cell proliferation(P<0.05).Conclusion:miR-455-3p inhibits cell proliferation in CRC,and its low expression is hopeful to be a potential biomarker for early diagnosis and prognosis assessment for CRC patients.
