Abstract:
Objective:To investigate the expression level and clinical significance of lymphatic enhancer binding factor 1(LEF1) in colon cancer patients.Methods:Expression level of LEF1 in tumor and adjacent tissues of colon cancer patients was observed by immunohistochemical staining combined with Western blot,and the correlation between LEF1 expression in tumor tissues and the clinicopathological characteristics of colon cancer patients was statistically analyzed.Kaplan-Meier survival analysis was used to analyze the effect of LEF1 expression level on the prognosis of colon cancer patients,and multivariate COX regression was used to analyze the risk factors affecting the prognosis of colon cancer patients.The expression level of LEF1 in cancer and adjacent tissues of colon cancer patients in the TCGA database was further analyzed,and the correlation between LEF1 expression and patient prognosis and lymphatic metastasis was analyzed.In addition,at the cellular level,Western blot was used to detect LEF1 protein expression in colon cancer cell lines,and the effect of LEF1 siRNA transfection on the biological function of colon cancer HCT116 cells was observed.Results:LEF1 expression level in tumor tissues of colon cancer patients was significantly increased compared with that in paracancer tissues(P<0.05),and its expression level was positively correlated with tumor pathological stage,degree of differentiation,and lymphatic metastasis(P<0.05),but not with patient age,pathological type(P>0.05).In addition,the survival time of colon cancer patients with high LEF1 expression levels was significantly lower than that of patients with low LEF1 expression levels(P<0.05).Multivariate COX regression analysis showed that high LEF1 expression level,tumor pathological stage,degree of tumor differentiation,and lymph node metastasis were risk factors for poor prognosis of colon cancer patients(P<0.05).The results of TCGA database analysis showed that the expression level of LEF1 in colon cancer tissues was significantly higher than that in adjacent tissues(P<0.05),and its expression level was negatively correlated with the prognosis of patients(P<0.05),and positively correlated with lymphatic metastasis of patients(P<0.05).In addition,the results of cell experiments showed that the expression level of LEF1 in human colon cancer cell lines was significantly higher than that in normal colon epithelial cells(P<0.05).LEF1 knockdown effectively reduced the migration and invasion abilities of colon cancer HCT116 cells and inhibited the activity of the Wnt/β-catenin signaling pathway(P<0.05).Conclusion:High LEF1 expression level is closely associated with low survival and poor prognosis of colon cancer patients and can be used as an important biomarker for postoperative recurrence,metastasis,and clinical prognosis of colon cancer patients.
