Abstract:
Objective:To investigate the effects of long non-coding RNA maternally expressed gene3(LncRNAs-MEG3) on the proliferation,migration and invasion of nasopharyngeal carcinoma(NPC) cells.Methods:Nasopharyngeal carcinoma cell lines TW03 and CNE-2 were cultured in vitro and transfected with MEG3,then they were divided into blank control group(NC),negative transfection group(negative control plasmid transfection) and experimental group(MEG3 mimis plasmid transfection).The content of LncRNAs-MEG3 in nasopharyngeal carcinoma cells was detected by real-time quantitative PCR,cell proliferation was detected by CCK-8,cell migration and invasion were detected by Transwell,and cell apoptosis was analyzed by flow cytometry.Results:The expression of MEG3 in NPC cells was significantly lower than that in NP69 cells(0.62±0.04)(P<0.05).Compared with the blank control group and the negative transfection group,the expression of MEG3 mRNA in the experimental group was increased(5.31±0.12)(F=441.062,P<0.01),and the proliferation,migration and invasion of nasopharyngeal carcinoma cells were decreased(F=10.832,P<0.05,F=1 534.134,P<0.05,F=1 430.212,P<0.05) and apoptosis increased(F=798.066,P<0.05).Conclusion:The expression of MEG3 in nasopharyngeal carcinoma is decreased,and the increase of MEG3 expression can inhibit the proliferation,migration,invasion of nasopharyngeal carcinoma cells,and promote cell apoptosis.The mechanism may be related to the inhibition of epithelial-mesenchymal transition.
