Abstract:
Objective:To explore the relationship between the long non-coding RNA HOXC antisense RNA1 (lncRNA HOXC-AS1),insulin like growth factor 2 mRNA binding protein 2 (IGF2BP2) mRNA expression levels in gastric cancer (GC) tissues and clinicopathological features and the survival of patients within 5 years after surgery.Methods:A total of 139 GC patients admitted to our hospital from May 2015 to April 2018 were selected.GC tissues and paracancerous tissues were collected surgically.The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression were detected by fluorescence quantitative PCR.GC patients were followed up for 5 years after operation.The survival of GC patients was recorded,and they were divided into survival group (88 cases) and death group (51 cases).The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues and paracancerous tissues,expression of lncRNA HOXC-AS1 and IGF2BP2 mRNA in GC tissues of patients with different clinical and pathological characteristics,clinicopathological features and lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels of GC tissues in survival group and death group were compared.The correlation between lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues,relationship between lncRNA HOXC-AS1 and IGF2BP2 mRNA expression in GC tissues and 5-year survival after surgery,risk factors for 5-year survival in patients with GC after surgery were analyzed.Results:The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues were significantly higher than those in paracancerous tissues (P<0.05).The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues were positively correlated (P<0.05).The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues were correlated with lymph node metastasis,TNM stage and tumor differentiation degree (P<0.05).The 5-year overall survival rate after surgery of lncRNA HOXC-AS1 high expression group and IGF2BP2 mRNA high expression group was significantly lower than that of lncRNA HOXC-AS1 low expression group and IGF2BP2 mRNA low expression group,respectively (P<0.05).There were significant differences in lymph node metastasis,TNM stage and tumor differentiation degree between survival group and death group (P<0.05).The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in GC tissues of death group were significantly higher than those of survival group (P<0.05).TNM stage III,poor tumor differentiation,high lncRNA HOXC-AS1 expression and high IGF2BP2 mRNA expression were independent risk factors affecting the survival of GC patients within 5 years after surgery (P<0.05).Conclusion:The lncRNA HOXC-AS1 and IGF2BP2 mRNA expression levels in cancer tissues of GC patients were significantly increased,and that in the death of GC patients within 5 years after surgery was higher than that of surviving patients.The two were closely related to lymph node metastasis,TNM stage and tumor differentiation degree,and their high expression was an independent risk factor affecting the survival of GC patients within 5 years after surgery.
