Abstract:
Objective:To investigate the value of abnormal expression of circRNA_0016418 and circRNA_0017247 in the diagnosis of malignant melanoma and the prediction of lymph node metastasis.Methods:The 68 patients with malignant melanoma in our hospital from January 2018 to October 2021 were selected and divided into lymph node metastasis group (n=26) and non-lymph node metastasis group (n=42).The circRNA_0016418 and circRNA_0017247 levels in malignant melanoma tissues and normal tissues were detected by real-time fluorescence quantitative PCR.The receiver operating characteristic (ROC) curve was used to analyze the value of abnormal expression of circRNA_0016418 and circRNA_0017247 in the diagnosis of malignant melanoma and its prediction of lymph node metastasis.Pearson correlation analysis detected the correlation between circRNA_0016418 level and circRNA_0017247 level in patients with malignant melanoma.Results:The levels of circRNA_0016418 (2.64±1.17 vs 0.95±0.36) and the levels of circRNA_0017247 (5.18±2.50 vs 1.52±0.63) in malignant melanoma tissues were significantly higher than those in normal tissues (P<0.001).The levels of circRNA_0016418 (3.85±1.72 vs 1.66±0.74) and the levels of circRNA_0017247 (7.16±3.82 vs 3.38±1.54) in the group with lymph node metastasis were significantly higher than those in the group without lymph node metastasis (P<0.001).ROC curve showed that the area under the curve of circRNA_0016418 and circRNA_0017247 for the combined diagnosis of malignant melanoma was 0.905 (95%CI:0.847~0.964),with sensitivity of 92.8% and specificity of 83.5%.The area under the curve of circRNA_0016418 combined with circRNA_0017247 to predict lymph node metastasis was 0.957 (95%CI:0.895~0.993),with sensitivity of 98.0% and specificity of 84.6%.Correlation analysis showed that circRNA_0016418 level was positively correlated with circRNA_0017247 level in patients with malignant melanoma (r=0.802,P<0.001).Conclusion:The levels of circRNA_0016418 and circRNA_0017247 in malignant melanoma tissues are significantly increased,which are associated with lymph node metastasis.The combined detection of the two is of great value in the diagnosis of malignant melanoma and the prediction of lymph node metastasis.
