Berbamine induces the apoptosis of gastric cancer AGS cells via endoplasmic reticulum stress pathway

Objective:To investigate the effects and the possible mechanisms of berbamine on the cell proliferation and apoptosis of human gastric cancer AGS cells.Methods:Cell viability was detected by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay.Cell apoptosis was detected by Annexin V/PI staining assay.The expression of Bcl-2,Bax,chop,GRP78 and cleaved caspase 12 proteins was detected by Western blot assay.Results:MTT assay result showed that berbamine could obviously inhibit the cell growth of AGS cells in a dosage-dependent manner.The cell apoptosis assay result showed that berbamine significantly increased the apoptotic rate of AGS cells.The Western blot result showed that berbamine down-regulated the expression of Bcl-2,while up-regulated the expression of Bax,chop,GRP78 and cleaved caspase 12 proteins.Moreover,compared with the berbamine group,the expression of chop in berbamine+chop siRNA group significantly decreased (P<0.01),while the expression of Bcl-2 in berbamine+chop siRNA group significantly increased (P<0.01).Conclusion:Berbamine could induce cell apoptosis via endoplasmic reticulum stress pathway in gastric cancer AGS cells.
