Abstract:
Objective:To investigate the effects of curcumin combined with KLF8-siRNA interference on proliferation,migration and apoptosis of lung cancer A549 cells and its mechanism.Methods:KLF8-siRNA interference sequence was transfected into A549 cells by liposome method,and the transfection effects were detected by RT-PCR and Western blot.The experiment was divided into NC group (negative control),KLF8-siRNA group (transfected KLF8-siRNA),Cur group (treated with 40 μmol/L curcumin) and KLF8-siRNA+Cur group (treated with curcumin after transfected KLF8-siRNA).The cell proliferation was detected by MTT and plate cloning.Cell migration was measured by Transwell chamber and scratch test.Cell apoptosis was detected by flow cytometry,and the expression of Wnt/β-catenin signaling pathway related proteins β-catenin,c-Myc,CyclinD1,MMP-2 and Survivin was detected by Western blot.Results:After transfection of KLF8-siRNA,the expression of KLF8 mRNA and protein in A549 cells decreased significantly.Compared with NC group,KLF8-siRNA transfection or curcumin treatment significantly reduced cell survival,clone formation,migration cell number and migration rate,while cell apoptosis rate significantly increased.Meanwhile,the expression levels of β-catenin,c-Myc,CyclinD1,MMP-2 and Survivin protein were significantly decreased (P<0.05).The above effects of curcumin combined with KLF8-siRNA interference on A549 cells were more significant than that of curcumin or KLF8-siRNA interference alone(P<0.05).Conclusion:Both KLF8-siRNA interference and curcumin can inhibit the proliferation,migration and promote apoptosis of A549 cells,and their combined effect is stronger.The mechanism may be related to the co-inhibition of Wnt/β-catenin signaling pathway activation.
