Abstract:
Objective:To study the expression and clinical significance of ubiquitin conjugating enzyme E2 O(UBE2O) in gastric cancer,clarify the effect of UBE2O on the proliferation and invasion of gastric cancer cells and explore the potential molecular mechanisms.Methods:The Cancer Genome Atlas(TCGA) database was used to analyze the expression differences of UBE2O mRNA in gastric cancer and normal tissues.Immunohistochemical staining was used to clarify the expression of UBE2O protein in gastric cancer and adjacent tissues.AGS and SGC-7901 cells were transfected with UBE2O shRNA.CCK-8 and transwell assays were used to detect cell proliferation and invasion ability.Western blotting assay was carried out to detect the expression of AMP-activated protein kinase α2(AMPKα2),phosphorylated mechanistic target of rapamycin(p-mTOR) and mTOR.Results:TCGA data indicated that the expression of UBE2O mRNA in gastric cancer tissues was significantly higher than that in normal tissues.Immunohistochemical staining results confirmed that the positive expression rate of UBE2O in gastric cancer tissues was prominently higher than that in adjacent tissues.UBE2O expression was significantly correlated with tumor size,depth of invasionand TNM stage.UBE2O knockdown markedly inhibited the proliferation and invasion of AGS and SGC-7901 cells.UEB2O silencing resulted in an increase in AMPKα2 protein level,while a decrease in p-mTOR protein level in AGS and SGC-7901 cells.Conclusion:UBE2O is up-regulated in gastric cancer tissues and is closely related to malignant clinicopathological characteristics.UBE2O may promote the proliferation and invasion of gastric cancer cells through AMPKα2/mTOR signaling pathway.
