Abstract:
Objective:To investigate the effect of long noncoding RNA(lncRNA) LINC-PINT/miR-425-5p/PTCH1 axis on laryngeal cancer stemness.Methods:The full-length sequences of LINC-PINT were amplified and transfected into HEp-2 cells using polymerase chain reaction(qRT-PCR) methods.Predict and verify the molecular targets of LINC-PINT and miR-425-5p using Diana Tools and dual luciferase report assay system respectively.qRT-PCR and Western blot were used to detect the expression of LINC-PINT and cancer stemness-associated genes(Sox-2,CD44,CD133 and Nanog) and their proteins.Results:After transfection of LINC-PINT,it was confirmed that the overexpression of LINC-PINT would cause the down-regulation of the expression of cancer stem-associated genes.Inhibition of miR-425-5p can down-regulate the expression of cancer stemness-associated genes.LINC-PINT regulated miR-425-5p and miR-425-5p targeted PTCH1.Conclusion:LINC-PINT may contribute to laryngeal carcinoma through inhibiting cancer cell stemness by miR-425-5p/PTCH1 axis.
