Abstract:
Objective:To investigate expression profiles of lactate dehydrogenase A/B (LDHA/LDHB) and their correlations with clinicopathological parameters of tumor in non-small cell lung cancer.Methods:Gene expression data and clinical information of non-small cell lung cancer,lung adenocarcinoma,and lung squamous cell carcinoma were obtained from GEO and TCGA databases.A nonparametric test was used to analyze the differential expression of LDHA/LDHB in cancer tissues and normal lung tissues.The survival of tumor patients was evaluated by Kaplan-Meier method and the Log-rank test.The chi-square test and Spearman's test were used to analyze the correlation between LDHA/LDHB expression and clinical features and tumor malignant biomarkers.The correlation between the expression of LDHA and LDHB and clinical features and tumor malignant biomarkers had also been elucidated.Results:LDHA and LDHB were highly expressed in non-small cell lung cancer,lung adenocarcinoma and lung squamous cell carcinoma.Lung adenocarcinoma patients with high expression of LDHA had shorter overall survival and progression-free survival.Lung adenocarcinoma patients with high expression of LDHB had shorter overall survival.Lung adenocarcinoma patients with high expression of LDHA and LDHB had the poorest overall survival and progression-free survival.The expression of LDHA and LDHB in patients with lung squamous cell carcinoma was not associated with survival status.In patients with lung adenocarcinoma,the expression of LDHA was involved in lymph node metastasis and pathological stage,and LDHB expression was associated with tumor stage.In patients with lung squamous cell carcinoma,LDHA expression was not associated with the clinical features,and LDHB was associated with age and lymph node metastasis.In lung adenocarcinoma,LDHA expression levels were positively correlated with PCNA,Ki67,and CDH2,and were not associated with CDH1.Expression levels of LDHB in lung adenocarcinoma were positively correlated with PCNA and Ki67,negatively correlated with CDH2,and not related to CDH1.Conclusion:The expression of LDHA and LDHB was associated with malignancy of lung adenocarcinoma.Elevated expression of LDHA and LDHB may indicate a poor prognosis and clinical features.
