Abstract:
Objective:To investigate the effect of Honokial and the possible mechanism on human cutaneous squamous cell carcinoma A431 cells.Methods:CCK8 assay was used to test the proliferation inhibitory effect of Honokial on cells.Quantitative Western blot was used to analyse the expressions of Bad and Bcl-2.Western blot was conducted to analyse the level of p53.Exogenous adenoviruses of p53 combined with Honokial were added to analyse the proliferation inhibitory and apoptosis effect on cells by Western blot assay.Results:Honokial could inhibit the growth of human cutaneous squamous cell carcinoma A431 cells in a time and concentration dependent way and induce apoptosis.Western blot assay showed Honokial up-regulated the level of p53.Western blot results showed that exogenous adenoviruses of p53 could enhance proliferation inhibitory and apoptosis effect induced by Honokial.Conclusion:Honokial could inhibit the proliferation and induce apoptosis on A431 cells,which may be mediated by p53 signal pathway.
