Abstract:
Objective:To investigate the molecular mechanism and interaction of MEMO1 and IGF-IR in the regulation of invasion and metastasis in gastric cancer cell lines,and to explore a potential target for clinical treatment in the gastric cancer.Methods:Five different differentiated gastric cancer cell lines (MKN-7,NCI-N87,MGC-803,BGC-823,MKN-74) were selected.Transwell was used to compare migration abilities of five gastric cancer cell lines.Western blot was used to detect the different expression levels of MEMO1,IGF-IR and E-cad.The expressions of EMT markers were detected by Western blot analysis after knockdown of MEMO1 or IGF-IR gene instantaneously in one of MEMO1 & IGF-IR over-expression cell lines,MGC-803.IP assay was used to detect the interaction between MEMO1 and IGF-IR.TCGA database was used to analyze the relationship between mRNA expression level of MEMO1 and IGF-IR and long-term survival in gastric cancer patients.Results:IGF-IR was significantly positively correlated with the expression of MEMO1.The migration abilities of five gastric cancer cells were MKN-7
