Abstract:
Objective:To study the effect of shRNA targeting silencing zinc finger E-box binding homeobox 2 (ZEB2) expression on the proliferation of lung cancer cells.Methods:shRNA-ZEB2 lentivirus and shRNA negative control lentivirus infected lung cancer cells.Real time PCR and Western blot were used to detected the silence effect.Cell proliferation activity was detected by MTT method,and cell apoptosis was detected by flow cytometry.Western blot was used to detect the levels of Cleaved Caspase-3,β-catenin and C-myc protein.Lung cancer cells silencing ZEB2 were disposed by Wnt/β-catenin signaling pathway inhibitor.Results:shRNA-ZEB2 lentivirus can obviously silence the expression and transcription of ZEB2 in lung cancer cells,but shRNA negative control lentivirus had no effect on the expression of ZEB2 in lung cancer cells.The proliferation activity of lung cancer cells was decreased after ZEB2 silencing.The rate of apoptosis was increased.The level of Cleaved Caspase-3 protein in the cells was increased,and the levels of β-catenin and C-myc protein were decreased.Wnt/ β-catenin signaling inhibitor downregulated the activation of Wnt/β-catenin signaling pathway in ZEB2 lung cancer cell lines.At the same time,cell survival can be reduced.Apoptosis was induced,and the expression of Cleaved Caspase-3 in the cell was promoted.Conclusion:shRNA targeting silencing ZEB2 expression reduces the proliferation activity of lung cancer cells through Wnt/β-catenin signaling pathway.
