Abstract:
Objective:To investigate the mechanism of CUL4A gene on the proliferation and invasion in gastric cancer.Methods:siRNA interference the expression of CUL4A in gastric cancer SCG-7901 cells.Using CCK-8 method to detect the proliferation of each group cells.To detect the invasion of each group cells by Transwell chambers.To detect each group cells apoptosis by Annexin V-FITC/PI method,and analysis the effects of CUL4A on cell apoptosis.Using Real time-PCR to analyze the expression of EMT related genes in gastric cancer cells.Results:siRNA interferenced the expression of CUL4A gene effectively in SCG-7901 gastric cancer cells(P<0.05).siRNA CUL4A inhibited the proliferation of gastric cancer cell(P<0.05).The results of Transwell chambers,interference CUL4A hindered the invasion of SCG-7901 cells(P<0.05).According to the results of cell apoptosis,siRNA CUL4A gene promoted the apoptosis of cells(P<0.05).Real time-PCR results showed that the expression of Snail mRNA and ZEB1 mRNA were lower than other group cells(P<0.05),the expression of E-cadherin mRNA was increased(P<0.05).Conclusion:CUL4A gene promoted gastric cancer cell proliferation,inhibited apoptosis and inhibition,and affected the expression of EMT related genes.
