Abstract:
Objective:To investigate the effect of salinomycin on radiosensitization of nasopharyngeal carcinoma CNE-2 cell line.Methods:CCK8 method was used to detect the effect of combination treatment with salinomycin and irradiation on proliferation.Plate clone assay was used to measure the clone formation rate.Hoechst33258 staining was used to perform morphological assessment of apoptotic cells.Cell cycle distribution and apoptosis were analyzed by flow cytometry.Results:CCK8 assay showed exposure to combination of salinomycin and irradiation resulted in decline of CNE-2 cell viability rate in concentration dose or dose-dependent manner and combination treatment had an additive inhibitory effect.Plate clone assay showed the combination treatment resulted in reduction of clone formation.In Hoechst33258 staining assay,apoptotic bodies of NPC cells added in response to the combination treatment.FCM assay showed the combination treatment induced strongly augmented number of apoptotic cells induced apoptosis and blocked cell cycle G1/S phase.Conclusion:Our study shows that salinomycin may enhance radiosensitization on CNE-2 cells.The radiosensitivity enhancement of may be related to apoptosis and cell cycle arrest.
