Abstract:
Objective:To assess the effect of FAK expression on apoptosis of human gastric carcinoma cells SGC-7901.Methods:The PGPU6/GFP/shNC(shNC)and PGPU6/GFP/FAK-299(shRNA-299)were designed and transfected into SGC-7901 cells.Interference effect of FAK was detected by Real-time PCR and Western blots.MTT assay was used to examine changes in cell proliferation.Cell apoptosis was analyzed by flow cytometry.The expression of associated apoptotic factors was measured by Western blots.Results:The expression of FAK in SGC-7901 cells significantly decreased in shRNA-299 group by contrast to the control group(P<0.05).Cells proliferation was inhibited by shRNA-299(P<0.05) and the level of cell apoptosis in shRNA-299 group was higher than in the control group(P<0.05).Western blots show an up-regulation of cleaved PARP,cleaved caspase-9,cleaved caspase-3,p53 and Bax,and a decrease in the expression of Bcl-2 protein in shRNA-299 group.Conclusion:FAK inhibition by shRNA-299 could activate apoptosis-associated proteins and induce apoptosis of human gastric carcinoma cells SGC-7901.
