Abstract:
Objective:To determine the distribution of curcumin and the characteristics of pharmacokinetics by high performance liquid chromatography-mass spectrometry (LC-MS/MS) in rat plasma and in vivo.Methods:Curcumin samples in rat plasma or tissue were collected and extracted by organic solvent ethyl acetate after oral administration.The method of high performance liquid chromatography-mass spectrometry with C18 chromatography column including acetonitrile (85%) and water (5%) as the mobile phase was used to analyzing the curcumin concentration in rat plasma or tissue.The mass spectrometry conditions were selected with electrospray ionization positive ion mode (ESI+) and multiple reaction monitoring (MRM) mode.Results:The peak time of curcumin was 0.5 in rat plasma and the AUC area was (236±6)ng?h/mL.In addition,the highest concentration of curcumin was (91±5)ng/mL in rat plasma.The peak time of curcumin was 1 in rat heart,but it was 2 in liver or kidneys,respectively.And the AUC area were (127±3)ng?h/mL and (166±5)ng?h/mL in the heart and liver,respectively.Conclusion:This LC-MS/MS method will be used to observe the characteristics of pharmacokinetics of curcumin rat plasma effectively.It was simple,highly reproducible and sensitive method.Curcumin could be absorbed into the blood,and distributed to the tissues widely after oral administration.
