Abstract:
Objective:To explore the effect of knockdown of SKA2 gene expression in human osteosarcoma cells on cell epithelial-mesenchymal transition (EMT),as well as the effect on cell proliferation,invasion and migration.Methods:Culture human osteosarcoma saos-2 and 143B cell lines.Then use the siRNA designed to synthesize the SKA2 gene and transfect the siRNA into saos-2 and 143B cells to form cells with low SKA2 expression.The mRNA level changes of SKA2 and EMT-related genes E-cadherin,Vimentin,Snail2 after siRNA transfection were detected by qRT-PCR method.Western blot method was used to detect the protein levels of SKA2,E-cadherin,Vimentin,and Snail2 after siRNA transfection.The Transwell experiment detects the change of cell invasion ability after the differential expression of SKA2 gene,the CCK-8 experiment detects the change of proliferation ability,and the scratch experiment detects the change of migration ability.Results:siRNA transfection established cells with low expression of SKA2 and reversed the EMT process.The expression of related genes Vimentin and Snail2 decreased,and the expression of E-cadherin increased.After siRNA transfection,the proliferation,invasion and migration of osteosarcoma cells were inhibited.Conclusion:SKA2 gene can mediates the invasion and metastasis of human osteosarcoma cells through the EMT process,and can affect the proliferation.
