Abstract:
Objective:To investigate the effect of inhibition of HIF-1α gene expression on the proliferation and apoptosis of hemangioma endothelial cells in vitro.Methods:The primary hemangioma cells were extracted by tissue block and enzyme digestion method,and the third generation cell was used in the experiment.Cells were divided into blank group,negative control group and HIF-1α transfection group.Expression of HIF-1α,Cleaved caspase3,Fas,PI3K,p-AKT protein after transfected 48 h was detected by Western bloting.CCK8 and flow cytometry were used to detect the proliferation and apoptosis of cells.Results:The expression of HIF-1ɑ in the negative control group and the blank group had no significant difference(P>0.05),while the expression of HIF-1α protein after transfected HIF-1α was significantly lower than the blank group(P<0.05).Cells survival rate in HIF-1α transfection group was decreased significantly,and the apoptosis rate was significantly increased,and the expression of Cleaved caspase3 and Fas protein had a significant increase,and PI3K and p-AKT protein expression were significantly decreased,and compared with the blank group,the differences were statistically significant(P<0.05).Conclusion:Inhibiting the expression of HIF-1α gene can decrease the proliferation of hemangioma endothelial cells and induce apoptosis,and the mechanism is related to down regulation of phosphatidylinositol 3-kinase/serine threonine kinase(PI3K/AKT) signaling pathway.
