Abstract:
Objective:To investigate the expression of lncRNA LINC01018 in NSCLC to evaluate its clinical significance,and study its influence in NSCLC.Methods:Total of 60 specimens of NSCLC tissues and normal lung tissues after surgery were collected.qRT-PCR was performed for LINC01018 expression in cancerous and normal lung tissues.The correlations between the expression of LINC01018 and pathological characteristics were analyzed.pcDNA-LINC01018 was transfected to up-regulate LINC01018 expression in NSCLC cells,and the transfection efficiency was evaluated by qRT-PCR.CCK8 assay was performed to evaluate the effect of ectopic LINC01018 expression on proliferation of NSCLC cells.The expression of RAC1 protein was determined by Western blot.Results:The expression level of LINC01018 was down-regulated in tumor tissue compared with the paired normal lung tissue(P<0.05).The expression of LINC01018 was associated with patients gender,tumor size,degree of differentiation,lymphatic metastasis and the distal metastasis (P<0.05).Meanwhile,over expression of LINC01018 reduced the expression of RAC1 and the proliferation of NSCLC cells were decreased.Conclusion:The expression of LINC01018 is obviously decreased in NSCLC patients with female patients,large tumor size,high clinical stage and advanced histological grade.LINC01018 may inhibit cell proliferation by affecting the expression of RAC1 proteins.
