Abstract:
Objective:To investigate the effects and mechanism of magnesium-L-threonate (L-TAMS) on the migration and invasion of non-small cell lung cancer H460 cells.Methods:Normal immortal lung epithelial cells 16HBE and non-small cell lung cancer cell line H460 were co-cultured with L-TAMS or magnesium sulfate (MgSO4).Intracellular magnesium concentration was detected by flow cytometry,cell migration ability was detected by scratches assay,cell invasion ability was detected by Transwell assay,and the expression of NF-κB/TNF-α pathway-related proteins was detected by Western blot,immunofluorescence staining (IF) and RT-qPCR.Results:Flow cytometry showed that the concentration of free magnesium ions in H460 cells decreased compared with that in the control group,and 0.4 mmol/L L-TAMS could supplement the concentration of free magnesium ions in H460 cells.Scratch test results showed that 0.4 mmol/L L-TAMS could significantly inhibit the migration of H460 cells,and the effect was better than that of 0.4 mmol/L MgSO4.The Transwell invasion experiment showed that L-TAMS inhibited the invasion ability of H460 cells.Western blot,IF,and RT-qPCR showed that L-TAMS could inhibit NF-κB/TNF-α pathway in a time-dependent manner,and the effect of 0.8 mmol/L L-TAMS was better than that of 0.4 mmol/L L-TAMS at 24 h.Conclusion:L-TAMS inhibited the invasion and migration and the NF-κB/TNF-α pathway of non-small cell lung cancer H460 cells,which provides a new way to search for cell drugs to prevent NSCLC in the clinic.
