Abstract:
Objective:To detect the expression level of miR-129-2 in cancer tissues of patients with triple-negative breast cancer(TNBC),analyze the relationship between the expression of miR-129-2 and clinical data of patients,and explore its influence on the migration and invasion of cancer cells.Methods:Real-time fluorescence quantitative PCR(RT-qPCR) was used to detect the expression level of miR-129-2 in TNBC cancer tissues,adjacent tissues and cell lines,and the correlation between the expression level of miR-129-2 and clinical data of patients was analyzed statistically.The effects of miR-129-2 on the migration and invasion of cancer cells were investigated by scratch assay and Transwell assay.Results:miR-129-2 expression was down-regulated in cancer tissues of 40 TNBC patients and MDA-MB-231 cells(P<0.05,P<0.01),and the expression level of miR-129-2 was not related to age and menopause,but was related to tumor size,stage and lymph node metastasis.The results of scratch test showed that overexpression of miR-129-2 reduced the migration distance of cells,the healing rate of scratches decreased,and the migration ability weakened(P<0.01).Transwell invasion assay showed that overexpression of miR-129-2 could reduce the number of invasion cells and significantly inhibit the invasion ability of cancer cells(P<0.01).Down-regulating the level of miR-129-2 enhanced the migration and invasion ability of MDA-MB-231 cells.Conclusion:The expression of miR-129-2 is low in TNBC,and the expression of miR-129-2 is related to the severity of the disease.The expression level of miR-129-2 affects the migration and invasion ability of TNBC cancer cells.
