Abstract:
Objective:To explore the effect of LncRNA RP11-214F16.8(Lnc-PCIR) on the proliferation and migration of laryngeal cancer cells by regulating the ubiquitination of poly(A)-binding protein cytoplasmic 4(PABPC4).Methods:Use Human Protein Atlas and GEPIA databases to analyze Lnc-PCIR and PABPC4 levels in head and neck squamous cell carcinoma tissues and the overall survival of patients.Select laryngeal cancer and adjacent tissue samples from 130 laryngeal cancer patients in our hospital,RT-qPCR and Western blot were used to detect Lnc-PCIR,PABPC4 mRNA and protein levels,and the relationship between Lnc-PCIR level and clinical pathological parameters of patients was analyzed.HEP-2,TU212 cells were transfected differently,as si-Lnc-NC group,si-Lnc-PCIR group,Lnc-NC group,Lnc-PCIR group,si-Lnc-PCIR+NC group,si-Lnc-PCIR+PABPC4 group.CCK-8,EdU stainingand Transwell test were used to detect the proliferation and migration ability.RT-qPCR was used to detect the Lnc-PCIR,PABPC4 mRNA levels.HEP-2 and TU212 cells were treated with cycloheximide(CHX),MG132 and ML364,Western blot was used to detect PABPC4 protein level.20 nudes mice were subcutaneously injected with transfected HEP-2 cell suspension and recorded as si-Lnc-NC group and si-Lnc-PCIR group.After one month,the volume,mass,and Lnc-PCIR and PABPC4 protein levels of the transplanted tumor were measured.Results:Database showed that Lnc-PCIR and PABPC4 levels in head and neck squamous cell carcinoma tissue are higher than those in normal tissue,and Lnc-PCIR and PABPC4 levels were negatively correlated with overall survival(all P<0.05).Compared with normal tissues,the mRNA and protein levels of Lnc-PCIR and PABPC4 were increased.Lnc-PCIR level was associated with gender,TNM stage,tumor differentiation,and lymph node metastasis(all P<0.05).After knocking down Lnc-PCIR,Lnc-PCIR and PABPC4 protein levels in cells were decreased.The OD value of culturing 1 d,2 d and 3 d,EdU positive rate and the number of cell migration were decreased.Overexpression of PABPC4 could partially reverse the effects of knocking down Lnc-PCIR on cell proliferation and migration(all P<0.05).Overexpression of Lnc-PCIR could reduce ubiquitination level of PABPC4(P<0.05).Knocking down Lnc-PCIR could inhibit the growth of transplanted tumors in mice(P<0.05).Conclusion:Lnc-PCIR promoted the proliferation and migration of laryngeal cancer cells by regulating the ubiquitination of PABPC4.
