Abstract:
Objective:To study the mechanism of miR-545-3p mediated by ultrasound microbubbles in inhibiting the malignant progression of breast cancer by regulating septin 2(SEPT2).Methods:The expression of miR-545-3p and SEPT2 in human normal breast epithelial cells MCF-10A,human breast cancer cells MCF-7,HS-578T and HCC-1937 cells were detected by real-time fluorescent quantitative PCR and Western blot.MCF-7 cells were cultured in vitro and randomly grouped into control group,empty microbubble group,miR-545-3p microbubble group,and miR-545-3p+SEPT2 overexpression microbubble group,after grouping and processing,real-time fluorescence quantitative PCR and immunoblotting experiments were applied to detect the expression of miR-545-3p and SEPT2 of cells in each group.CCK-8,colony formation assay,and flow cytometry were applied to detect cell proliferation and apoptosis in each group.Cell scratch and Transwell invasion tests were applied to detect cell migration and invasion in each group.Nude mice were subcutaneously inoculated with MCF-7 cells on the abdominal wall to construct their transplanted tumor model and grouped using the same method.After grouping,the tumor volume and weight of each group of nude mice were measured.Results:Compared with MCF-10A cells,the expression of miR-545-3p in MCF-7,HS-578T,and HCC-1937 cells decreased(P<0.05),the expression of SEPT2 mRNA and protein increased(P<0.05).Compared with the control group,the miR-545-3p expression and apoptosis rate in the miR-545-3p microbubble group increased(P<0.05),the cell SEPT2 mRNA and protein expression,cell viability,colony formation rate,migration rate,invasion number,and tumor volume and weight in nude mice reduced(P<0.05).Compared with the miR-545-3p microbubble group,the cell apoptosis rate in the miR-545-3p+SEPT2 overexpression microbubble group decreased(P<0.05),the cell SEPT2 mRNA and protein expression,cell viability,colony formation rate,migration rate,invasion number,and tumor volume and weight in nude mice increased(P<0.05).Conclusion:Ultrasound microbubble-mediated miR-545-3p can reduce the proliferation,migration and invasion of breast cancer cells,promote their apoptosis,inhibit the growth of transplanted tumor in nude mice,and finally inhibit the malignant progression of breast cancer cells by reducing the expression of SEPT2.
