Abstract:
Objective:To investigated the expression of CPNE7 in gastric cancer tissues and cells and its biological effects on the proliferation,migration and invasion of gastric cancer cells.Methods:The differential expression of CPNE7 in gastric cancer and adjacent normal tissues was analyzed at the transcriptional level based on the bioinformatics database GEPIA.The expression of CPNE7 was detected by qRT-PCR in three gastric cancer cell lines(AGS,MKN-45,HGC-27) and normal gastric epithelial cells(GES-1),and AGS cells with relatively high expression levels were selected as subsequent experimental cells.The expression of CPNE7 was knocked down by lentiviral transfection of AGS cells and the cells divided into RNAi-vector group,RNAi-1 group,RNAi-2 group,and RNAi-3 group according to different treatments,and the transfection efficiency was verified by qRT-PCR and Western blot,and RNAi-1(experimental group) and RNAi-vector(control group),which had the most effective knockdown efficiency,were selected for subsequent experiments.CCK-8 assay was used to detect cell viability.EdU and colony formation assay were used to detect cell proliferation and colony formation ability.Scratch healing assay and Transwell assay were used to detect cell migration and invasion ability,and Western blot was used to detect the expression levels of apoptosis-related proteins(Caspase3,Caspase7,Caspase9,Bax,Bcl-2) and epithelial-mesenchymal transition(EMT)-related proteins(E-cadherin,N-cadherin,Vimentin).Results:The expression of CPNE7 in gastric cancer tissues was significantly higher than that in normal tissues(P<0.05).Compared with normal gastric epithelial cells(GES-1),CPNE7 was highly expressed in gastric cancer cells(P<0.05),and the relative expression of CPNE7 was the highest in AGS gastric cancer cells(P<0.001).Compared with the RNAi-vector group,knockdown of CPNE7 inhibited the proliferation,migration and invasion of gastric cancer cells(P<0.05).Compared with the RNAi-vector group,knockdown of CPNE7 up-regulated the expression of apoptosis-related proteins Caspase3,Caspase7,Caspase9,and Bax and down-regulated the expression of Bcl-2(P<0.05).Compared with the RNAi-vector group,knockdown of CPNE7 increased the expression level of epithelial marker proteins(E-cadherin)(P<0.01) and decreased the expression level of mesenchymal marker proteins(N-cadherin,Vimentin)(P<0.01).Conclusion:CPNE7 is up-regulated in gastric cancer tissues and cells,and knockdown of CPNE7 can inhibit gastric cancer cell proliferation,migration and invasion by affecting the EMT process,which may become a potential molecular marker of gastric cancer.
