Abstract:
Objective:To investigate the expression of protein kinase D3 (PRKD3) in colorectal cancer tissue and its clinical significance.Methods:Bioinformatics was used to analyze the expression of PRKD3 and its correlation with clinicopathological parameters.The protein expression of PRKD3 in 81 cases of colorectal cancer tissue and paired adjacent normal tissue was detected by tissue microarray and immunohistochemical staining.The correlation between the protein expression of PRKD3 and the clinicopathological parameters as well as the prognosis of the patients was analyzed.qPCR was used to detect the expression of PRKD3 mRNA in 8 cases of colorectal cancer tissue and paired adjacent normal tissue.Results:Bioinformatics analysis showed that the expression level of PRKD3 mRNA in colorectal cancer tissue was significantly lower than that in normal colorectal tissue (P<0.05),and its expression was significantly correlated with lymphatic vessel invasion and the history of colon polyps (P<0.05).Immunohistochemical staining showed that PRKD3 protein was mainly located in the cytoplasm of colorectal cells.The positive expression rate of PRKD3 in adjacent normal tissue was 26.4% (19/72),and that in colorectal cancer tissue was 63.0% (51/81).There was a statistical difference between the two groups (P<0.05).qPCR results showed that PRKD3 mRNA expression in adjacent normal tissue was significantly higher than that in colorectal cancer tissue.The protein expression level of PRKD3 was correlated with the clinical diagnosis of colorectal cancer (P<0.05).Kaplan-Meier survival curve showed that the postoperative survival time of patients with high PRKD3 expression was significantly longer than that of patients with low PRKD3 expression (P<0.05).Multivariate COX regression analysis showed that the high expression of PRKD3 was not an independent factor for the prognosis of colorectal cancer patients.Conclusion:PRKD3 may be involved in the occurrence and development of colorectal cancer,and it can be used as a potential molecular marker for the prognosis and treatment of colorectal cancer patients.
