Abstract:
Objective:To investigate the effect of diallyl disulfide (DADS) down-regulation of DJ-1 on the invasion and epithelial-mesenchymal transformation of human gastric cancer MGC803 cells through PTEN/Akt pathway.Methods:The experiment was divided into MGC803 group,empty carrier group,DJ-1 overexpression group,MGC803+DADS group,empty carrier+DADS group,DJ-1 overexpression+DADS group.The effects of DADS on proliferation,clonal formation,migration and invasion of MGC803 cells were determined by MTT,plate cloning,cell scratch and invasion assay.The effect of DADS on the morphology of MGC803 cells was observed by contrast microscope.The expressions of DJ-1,PTEN,p-Akt,Snail,Vimentin,E-cadherin,MMP-9 and TIMP-3 were detected by qRT-PCR,Western blot and immunofluorescence.Results:MTT showed that after 24 h,48 h and 72 h, the proliferation rate of DADS treatment in all groups decreased compared with that before treatment (P<0.05).Plate cloning showed that the number of clones formed after DADS treatment was lower than that before DADS treatment (P<0.05).The scratch test showed that 24 h after DADS treatment,the scratch distance increased compared with that before treatment (P<0.05).Migration experiment showed that after DADS treatment,the number of transplanted cells in all groups decreased compared with those before DADS treatment (P<0.05).Invasion experiment showed that DADS invaded less cells after treatment than before treatment (P<0.05).DJ-1 expression decreased after DADS treatment (P<0.05),PTEN expression increased after DADS treatment (P<0.05),and immunofluorescence was consistent with the above results.After DADS treatment,the expression of p-Akt was down-regulated in all groups (P<0.05).Phase contrast microscopy showed that after DADS treatment,the deformability of MGC803 group and DJ-1 overexpression group decreased,Snail,Vimentin and MMP-9 expression were down-regulated,and E-cadherin and TIMP-3 expression were up-regulated (P<0.05).Conclusion:DADS down-regulation of DJ-1 can negatively regulate PTEN/Akt pathway and inhibit the invasion and epithelial-mesenchymal transformation of human gastric cancer MGC803 cells.
