Abstract:
Objective:To detect the expression of MDA-9 in colorectal cancer cells,and determine its role on migration and invasion of SW620 cells.Methods:The mRNA and protein expression levels of MDA-9 in normal colorectal epithelial cell line (NCM460) and three colorectal cancer cell lines (HCT116,SW480 and SW620) were determined by RT-qPCR and Western blot assay.The small interfering RNA targeting MDA-9 (si-MDA-9) and negative control (si-NC) were transfected into SW620 cells,and scratch healing assay and Transwell assay were used to detect the effect on cell migration and invasion.The proteins expression of p-FAK and MMP-2 was determined by Western blot assay.Results:MDA-9 mRNA expression level (P<0.001) and protein expression level (P<0.01) were significantly up-regulated in three colorectal cancer cell lines than in NCM460 cells.The relative migration ability and invasion ability of SW620 cells in the si-MDA-9 group were (60.7± 5.8)% and (55.3±10.3)%,which were lower than (98.3±14.3)% and (98.4±9.5)% in the si-NC group,and (100±9.8)% and (100±13.1)% in the blank control group (P<0.01).The protein levels of p-FAK (P<0.05) and MMP-2 (P<0.01) in SW620 cells of si-MDA-9 group were also lower than those of si-NC group and blank control group.Conclusion:MDA-9 is highly expressed in colorectal cancer cells,and inhibiting its expression level will lead to a decline in the migration and invasion ability of SW620 cells,which may be related to inhibiting the expression of p-FAK and MMP-2 proteins.
