Abstract:
Objective:To explore the effect of PUM1 gene on proliferation,migration and invasion ability of glioma cells.Methods:siRNA interfered with the expression of PUM1 in U373 and U251 cells.The expressions of PUM1 mRNA in U373 and U251 cell lines were detected using RT-qPCR.CCK-8 assay was employed to assess the proliferative capacity of U373 and U251 cells pre- and post-siRNA-mediated interference of PUM1.The apoptosis of U373 and U251 cells was assessed by TUNEL assay before and after interference with siRNA-PUM1.Cell scratch assay was used to evaluate the migration ability of U373 and U251 cells before and after PUM1 siRNA interference.The invasion ability of U373 and U251 cells was assessed using Transwell before and after interference with siRNA-PUM1.The lactate assay kit was utilized to assess the alterations in lactate expression pre- and post-PUM1 silencing in U373 and U251 cells using siRNA.Western blotting detected the expression changes of PKM2 and Parkin in U373 and U251 cells before and after siRNA-PUM1 interference.Results:siRNA-PUM1 significantly decreased the expression of PUM1 in U373 and U251 cells.siRNA-PUM1 significantly suppresses the proliferation,migration,and invasive capacity of U373 and U251 cells.siRNA-PUM1 significantly suppresses glycolytic activity in U373 and U251 cells,as well as the expression of PKM2 and Parkin.Conclusion:Silencing of the PUM1 gene using siRNA may potentially impede the proliferation,migration,and invasion capabilities of U373 and U251 glioma cells by suppressing the glycolytic pathway.
