Abstract:
Objective:In order to improve the diagnostic accuracy of patients with epithelial ovarian cancer (EOC) in clinical practice,the expression and clinical significance of microRNA-5766-3p (miR-576-3p) and microRNA-766-5p (miR-766-5p) in the serum of EOC patients were investigated.Methods:A total of 103 EOC patients admitted to our hospital from August 2017 to August 2022 were regarded as the study group,103 patients with ovarian cysts were selected as the benign control group,and another 103 healthy individuals who underwent physical examination at our hospital during the same period were selected as the healthy control group.Measure and analyze the expression levels and correlation of serum miR-576-3p and miR-766-5p in patients,and explore the influencing factors related to the occurrence of EOC.Evaluate the diagnostic value of miR-576-3p and miR-766-5p for EOC using receiver operating characteristic curve (ROC).Results:There was a significant difference in PLR and NLR values among the three groups(χ2=151.671,69.059,P<0.05).The serum miR-576-3p expression level in the study group (0.76±0.22) was lower than that in the healthy control group (1.02±0.17) and benign control group (0.92±0.12) (F=57.959,P<0.001),while the miR-766-5p expression level in the study group (1.24±0.45) was significantly higher than that in the healthy control group (1.01±0.11) and benign control group (1.11±0.31) (F=13.227,P<0.001),and the serum miR-576-3p expression level increased (0.89±0.11) (t=5.364,P<0.001) after treatment,and the expression level of serum miR-766-5p decreased (1.11±0.35) (t=2.314,P<0.05).The serum miR-576-3p expression levels in EOC patients with FIGO stage III+IV (0.58±0.16),lymph node metastasis (0.60±0.18),and poor degree of differentiation (0.63±0.20) were lower than those in EOC patients with FIGO stage I+II (0.87±0.26),no lymph node metastasis (0.85±0.35),and well degree of differentiation (0.82±0.31) (P<0.05),while the trend of serum miR-766-5p expression levels was opposite.PLR (OR=2.114),NLR (OR=1.594),and miR-766-5p (OR=1.730) were all risk factors for EOC,while miR-576-3p (OR=0.627) was a protective factor (P<0.05).The area under the curve (AUC) of serum miR-576-3p,miR-766-5p,PLR,NLR,and combined detection for diagnosing EOC was 0.801 (95%CI=0.739~0.853),0.741 (95%CI=0.676~0.800),0.906 (95%CI=0.857~0.942),0.776 (95%CI=0.713~0.831),and 0.970 (95%CI=0.937~0.989),respectively.Joint detection was superior to individual detection (Zcombined vs miR-576-3p=6.149,Zcombined vs miR-766-5p=6.300,Zcombined vs PLR=3.550,Zcombined vs NLR=5.592,P<0.001).Conclusion:There is a certain correlation between the expression levels of serum miR-576-3p and miR-766-5p and the occurrence of EOC.The combined detection has high sensitivity in diagnosing EOC and has certain application value in assisting clinical diagnosis of EOC.
